The Recombigen Cholesterol Direct (EP), HDL assay uses a two-reagent system designed for the direct measurement of total cholesterol and HDL cholesterol in serum or plasma samples. The assay is based on the CHOD-POD (Cholesterol Oxidase-Peroxidase) method for total cholesterol, along with phosphotungstate for HDL cholesterol. The method provides accurate and reliable results for lipid profiling, which is critical for cardiovascular risk assessment.

In the first reagent, CHOD (Cholesterol Oxidase) reacts with cholesterol to produce cholest-4-en-3-one and hydrogen peroxide (H₂O₂). In the second reagent, peroxidase (POD) reacts with H₂O₂ to form a colored complex, which is measured spectrophotometrically. The specific interaction with phosphotungstate selectively precipitates the HDL fraction, allowing for the quantification of HDL cholesterol separately from total cholesterol.
