Dosage and Administration

  1. Leeporate Injection is for intravenous use only and must be administered by a healthcare professional.
  2. Dosage is highly individualised, based on age, body weight, and clinical response.
  3. It can be given as a slow intravenous injection over 3-5 minutes or by continuous or repeated infusion.
  4. The injection should be diluted with compatible intravenous solutions such as normal saline, dextrose 5%, or dextrose saline.
  5. It should be replaced by oral valproate therapy as soon as clinically feasible.

 

Storage and Disposal

  • Store vials at controlled room temperature, typically between 15-30°C (59-86°F) or 20-25°C (68-77°F).
  • Protect from light and moisture.
  • Keep out of reach of children.
  • Discard any unused portion of the 5ml vial after opening.

Precautions and Safety

  • Sodium Valproate carries a risk of severe hepatotoxicity (liver damage), especially in children under two years of age, and those with congenital metabolic disorders or on multiple anticonvulsants. Liver function tests should be monitored frequently, particularly during the first six months of treatment.
  • Pancreatitis, which can be severe and fatal, has been reported. Patients experiencing symptoms like abdominal pain, nausea, or vomiting should seek immediate medical evaluation.
  • Valproate has a high teratogenic potential, posing significant risks of congenital malformations and neurodevelopmental disorders if used during pregnancy. Women of childbearing potential must use effective contraception and be enrolled in a pregnancy prevention program.
  • Hyperammonemic encephalopathy may occur. Monitor for unexplained lethargy, vomiting, or changes in mental status, and measure ammonia levels if suspected.
  • Avoid concomitant use with carbapenem agents due to potential drug interactions.
  • This medicine may cause drowsiness, dizziness, or blurred vision; avoid driving or operating machinery until you know how it affects you. Alcohol can intensify these effects.
  • Contraindicated in patients with known urea cycle disorders or mitochondrial disorders.

 

Side Effects and Risks

  • Common side effects include nausea (especially after intravenous injection), vomiting, abdominal pain, headache, dizziness, and drowsiness.
  • Other possible side effects include tremors, changes in vision or taste, irritation at the injection site, and unusual bruising or bleeding.
  • Serious but rare side effects include severe skin reactions, allergic reactions, and hypothermia (low body temperature).
